Deregulations of RNA Pol II Subunits in Cancer

چکیده

Deregulated transcription is a well-known characteristic of cancer cells, with differentially expressed genes being common feature several cancers. Often, deregulated consequence alterations in factors (TFs), which play crucial role gene expression and can act as tumour suppressors or proto-oncogenes. In eukaryotic organisms, carried out by three distinct RNA polymerase complexes: Pol I, II, III. specifically, responsible for transcribing messenger (mRNA), the protein coding part genome, well long non-coding RNAs (lncRNAs). While there considerable research on impact specific development, lack studies focusing defects within complexes their subunits. This review aims to shed light particular II complex highlight deregulation its subunits that have significant prognosis, survival. By providing comprehensive overview our current understanding cancer, this emphasizes importance further area. It suggests exploring these subunits’ deregulations could lead identification valuable biomarkers potential therapeutic targets, making it topic collective interest.

A myosin for RNA Pol II

A myosin for RNA Pol II T ranscription gets a boost from myosin VI, according to fi ndings from Sarah Vreugde (DIBIT Scientifi c Institute, Milano, Italy) and colleagues. The motor might drag genes into transcription neighborhoods. While trying to understand the function of myosin VI, Vreugde noticed that its localization pattern looked much like that of RNA polymerase II. The authors then show...

Epigenetic deregulations in gastric cancer.

Abnormal functioning of many cellular processes such as cell cycle, DNA repair, angiogenesis and cell-cell adhesion has been reported in all types of cancer. In the context of cancer, genes responsible to sustain integrity of the cells can be categorized into tumor suppressor genes and oncogenes. In normal conditions, these genes maintain a state of equilibrium and imbalance of these two groups...

CIITA regulates transcription onset viaSer5-phosphorylation of RNA Pol II.

We describe the temporal order of recruitment of transcription factors, cofactors and basal transcriptional components and the consequent biochemical events that lead to activation of the major histocompatibility class II (MHCII) DRA gene transcription by IFN-gamma. We found that the gene is 'poised' for activation since both the activators and a fraction of the basal transcriptional machinery ...

Cdc48/p97 mediates UV-dependent turnover of RNA Pol II.

Cdc48/p97 is an essential ATPase whose role in targeting substrates to the ubiquitin-proteasome system (UPS) remains unclear. Existing models posit that Cdc48 acts upstream of UPS receptors. To address this hypothesis, we examined the association of ubiquitin (Ub) conjugates with 26S proteasomes. Unexpectedly, proteasomes isolated from cdc48 mutants contain high levels of Ub conjugates, and mas...

E2F1-dependent methyl cap formation requires RNA pol II phosphorylation

Gene expression is a process integral to cell proliferation. The E2F family of transcription factors upregulates expression of transcripts whose products are essential for cell cycle progression. Here, we report that E2F1 promotes gene expression by an additional mechanism, that is, formation of the methyl cap on RNA pol II transcripts. The methyl cap is required for mRNA maturation, expression...
